How can I fix sound on HTC Desire without changing the amplifier or replacing the phone?

The phone is otherwise working properly except that it has lost ability to produce sound of any kind.

How to fix sound on HTC Desire without changing amplifier.

It has found that there is system error in HTC causing towards poor sounds. This is caused by most likely through a system bug in android. I guess HTC Company is still working on this issue. There is no use of doing hard resets or formatting. But there may be two issues on your phone.

1. If the audio problem has caused due to installing a 3rd party application or software (many of those applications do have the ability to control system sounds), you will have to manually uninstall it.

2. If this is a system error, there is an application called "AudioManager", which allows you to use more settings of the volume of the phone than the control panel settings. Do a Google search on the software and install it after downloading. In most of the times it will fix the error in audio of your phone.
